Hedgehog是一種共價(jià)結(jié)合膽固醇的分泌性蛋白,在動(dòng)物發(fā)育中起重要作用。果蠅的該基因突變導(dǎo)致幼蟲(chóng)體表出現(xiàn)許多刺突,形似刺猬,故名Hedgehog。脊椎動(dòng)物中至少有3個(gè)基因編碼Hedgehog蛋白,即:Shh(Sonic hedgehog)、Ihh(Indian hedgehog)和Dhh(desert hedgehog)。
Hedgehog(Hh)信號(hào)通路調(diào)節(jié)胚胎發(fā)育和成體許多組織器官干細(xì)胞的自我更新與增殖。然而,那些在正常發(fā)育過(guò)程中受到Hh信號(hào)通路調(diào)節(jié)的組織器官,在該信號(hào)通路異常時(shí)常常發(fā)生腫瘤。這些腫瘤包括肝癌、神經(jīng)膠質(zhì)瘤、基底細(xì)胞癌、橫紋肌肉瘤、胰腺癌、小細(xì)胞肺癌、胃癌、結(jié)腸癌、前列腺癌、黑色素瘤和多發(fā)性骨髓瘤等。

Hedgehog基因zui早在果蠅中作為體節(jié)極性基因被發(fā)現(xiàn),Hedgehog基因編碼一種分泌信號(hào)蛋白,突變會(huì)使無(wú)毛部分變成有毛部分,所以又戲稱為“刺猬"基因。哺乳動(dòng)物發(fā)現(xiàn)其同源基因有3個(gè),分別為Sonic Hedgehog(SHh)、Indian Hedgehog(IHh)和Desert Hedgehog(DHh)。SHh 前體蛋白要經(jīng)過(guò)一系列剪切與修飾,首先SHh 前體蛋白在內(nèi)質(zhì)網(wǎng)剪切去掉信號(hào)肽,,然后經(jīng)過(guò)自我酶解形成19 kDa N 端蛋白和25 kDaC 端蛋白,SHh-N 端蛋白發(fā)揮了SHh信號(hào)分子的功能,執(zhí)行SHh信號(hào)通路的主要功能。C 端蛋白擁有末端膽固醇轉(zhuǎn)移酶活性,可以將膽固醇分子轉(zhuǎn)移到SHh N 端蛋白的C 端,末端的膽固醇分子可以和細(xì)胞膜結(jié)合而錨定SHh N 端蛋白,從而有效控制SHh N 端蛋白的作用區(qū)域。
經(jīng)典的SHh信號(hào)通路是由SHh配體、2個(gè)跨膜蛋白質(zhì)收徒Patched(Ptch)和 Smoothened(Smo)組成的受體復(fù)合物,以及下游的轉(zhuǎn)錄因子Gli蛋白(Gli-1、Gli-2和Gli-3)組成。
Hedgehog信號(hào)在胚胎發(fā)生過(guò)程中細(xì)胞分化和器官形成起到非常重要的作用,在成體一些組織器官Hh信號(hào)通路仍然活躍,調(diào)節(jié)干細(xì)胞靜止與增殖,Hh基因在不同組織過(guò)度表達(dá)或靶基因突變等,均可導(dǎo)致細(xì)胞特異性增生,參與腫瘤的發(fā)生與發(fā)展。

The hedgehog (Hh) gene encodes a secreted protein originally identified in Drosophila. The vertebrate homologs of Hh comprise of Sonic (SHh), Desert (DHh) and Indian (IHh). SHh plays an important role in embryonic development and in diseases such as cancer. Human SHh precursor is a 45kDa protein. An autocatalytic reaction yields a 19 kDa N-terminal domain that is biologically active. Because of unique post-translational modifications, SHh produced in mammalian cells is much more biologically active than SHh produced from E. coli and is devoid of artifact associated with the E. coli products commercially available from other manufacturers.
